| 内容記述 | 目的：Transient receptor potential melastain (TRPM) subfamilyは，中枢および末梢神経系において温度受容や神経組織形成に寄与する．しかし，末梢神経系の髄鞘を構成するシュワン細胞におけるTRPM subfamilyの解析は進んでいない．そこで，ラット後根神経節より樹立された成熟シュワン細胞株immortalized Fischer rat Schwann（IFRS）1細胞におけるTRPM subfamilyの発現パターンを解析した． 方法：IFRS1細胞より総RNAを抽出後，RT-PCRに付すことで各TRPM subfamily 遺伝子の発現パターンを確認した．また，IFRS1細胞からタンパク質を調製し，ウェスタンブロットにより各TRPM subfamilyのタンパク質発現を確認した． 結果：RT-PCRによりTRPM3，M4，M7，M8 遺伝子の発現が確認されたが，TRPM1，M2，M5，M6 遺伝子の発現は認められなかった．さらに，遺伝子発現が確認された4つのTRPM subfamilyは，ウェスタンブロットによりタンパク質発現も認められた． 考察：TRPM3，M4，M8は温度感受性チャネルであり，TRPM7は髄鞘形成に関与することが報告されている．IFRS1細胞は，TRPM subfamilyに起因する温度過敏や脱髄などの末梢神経異常を調べるために応用できる可能性が示唆された． |

| 内容記述 | Purpose: Transient receptor potential melastatin (TRPM) subfamily is a non-selective cation channel, playing a key role in physiological functions and diseases of the central nervous system. These channels affect the peripheral nervous system (PNS); however, the expression patterns of the TRPM subfamily are unknown in Schwann cells that constitute the myelin sheath of the PNS. We analyzed the TRPM subfamily expression in immortalized Fischer rat Schwann cells 1 (IFRS1) as mature Schwann cells. Methods: We analyzed the expression of TRPM subfamily genes in IFRS1 cells using reverse transcription-polymerase chain reaction (RT-PCR). Additionally, the protein expression of TRPM3, TRPM4, TRPM7, and TRPM8 was examined by western blotting. Results: We confirmed the expression of TRPM3, TRPM4, TRPM7, and TRPM8 in IFRS1 cells. These channels were also expressed at the protein level. Conclusion: We found the expression of TRPM subfamily (i.e., TRPM3, TRPM4, TRPM7, and TRPM8) in immortalized adult rat Schwann cells, IFRS1. Therefore, we suggest IFRS1 cells could be one of the tools used for the evaluation of peripheral nerve abnormalities, such as demyelination and temperature hypersensitivity, involved with the TRPM subfamily |
